Background:The present study evaluated the effect of the current treatment of erectile dysfunction (ED) on the plasma concentrations of sildenafil (Viagra), tadalafil (Cialis) and avanafil (Avanafil) in healthy male subjects.
Method:Twenty-four healthy male subjects aged between 20 and 40 years, with ED of about 6-8 mm Hg and a body weight of more than 30 kg, were recruited and randomly administered 1 g of sildenafil, tadalafil and avanafil orally once daily, for 3 days. Blood samples were obtained before, during and after the treatment. Serum samples were analyzed for plasma concentration of sildenafil, tadalafil and avanafil. Blood sampling was performed on the same days of the study. Plasma samples were obtained by centrifugation of ED plasma at 3,500 rpm for 30 min and then the supernatant was used for quantification of sildenafil, tadalafil and avanafil. Blood sampling was performed after the administration of plasma samples.
Results:Following administration of sildenafil, tadalafil and avanafil, a statistically significant increase in plasma sildenafil was observed. On the other hand, tadalafil and avanafil did not exhibit any significant effect.
Conclusion:Sildenafil, tadalafil and avanafil were found to be the active metabolite of sildenafil and tadalafil and avanafil is a strong metabolite of avanafil.
